Description
Lentivirus express TCR (a-GalCer) (Clone NTK15), containing no any antibiotic selection. This TCR actives the T-Cells with the CD1D lipid Antigen-activated antibody recognizing galactosylceramide (GalCer) (Clone (NKT15). It is often used to engineer invariant natural killer T (iNKT) cells
The TCR construct express the antibody specifically recognizes the lipid-based antigen, α-galactosylceramide (α-GalCer) (presented by CD1d). The TCR α chain variable region with a transmembrane (TM) domain and β chain variable region with a transmembrane (TM) domain, are linked by F2A element, as “TCRα(V)/Tm-F2A-TCRβ(V)/Tm”
The “target-specific TCRαV and TCRβV sequences” are derived from the verified clones according to published literatures. It also transduces a GFP–Puromycin (fluorescent-antibiotic fusion ) dual selection gene under the separate RSV promoter, which allow to enrich or select the transduced cells via GFP sorting or Puromycin killing. see the Lentivector scheme below..
About α-Galactosylceramide (α-GalCer):
α-GalCer is a glycolipid antigen that can bind to CD1d molecules. α-GalCer is a potent activator of invariant NKT (iNKT) cells. When α-GalCer is presented by CD1d, it cells activates NKT cells, a unique subset of T cells that bridge the innate and adaptive immune responses. α-GalCer has been utilized extensively due to its ability to induce potent activation of mouse and human iNKT cells. Amount: 200 ul/vial at titer of 1×108 IFU/ml, concentrated lentivirus provided in PBS solution, premixed with 10x polybrene (60ug/ml), premade, ready to ship in dry-ice package.
