Control lentivirus
Gentarget provides the corresponding control lentivirus for over-expression, inducible expression, shRNA knockdown. They have the same lentivector backbone as the matched target lentivirus, but cloned a Null-control sequence. The control lentivirus serves as the serves as the negative controls for lentivirus treatment, or validates the specificity of any target expression.
1. Target over expression control lentivirus:
The negative control lentivirus for gene over-expression, cloned a Null spacer sequence at the target position under the engineered CMV promoter or the enhanced EF1a promoter. Due to the lack of a start codon, the Null sequence does not express a target but still contains the antibiotic resistance under the RSV promoter (except the product without any selection). See the control lentivirus’ core structure in the scheme below:
2. shRNA expression control lentivirus:
The negative control shRNA lentivirus contains an universal negative control shRNA sequence under either the optional inducible H1 promoter or constitutive U6 promoter. It validates the off-target effects of shRNA treatment. See control shRNA lentivector map scheme below.
Please select the control lentivirus (see product list below), with matched promoter and antibiotic selection as your target shRNA lentivirus.
3. Optional inducible expression control lentivirus:
GenTarget provides pre-made Inducible expression lentivirus of Fluorescent report, which can be used to validate the optional inducible expression system in your specific cell lines.
The inducible controls contain GFP or RFP under control of the inducible promoter, enabling very convenient verification of tetracycline inducible effects by simple monitoring of GFP or RFP signal under a microscope. The controls also contain a fluorescent-antibiotic fusion dual marker under a RSV promoter which is not affected by induction. The dual marker provides a convenient way to monitor viral transduction efficiency and select positive cells by either antibiotic killing or fluorescence sorting. see the core lentivector scheme below:
