Unstable GFP
About Unstable GFP (destabilized GFP) :
The green fluorescent protein (GFP) is a widely used reporter, providing an easy detection in living cells. However, it is a very stable protein and accumulated in cells with long half-live, which limits its application that requires fast turnover responses in signal pathway assay and in knockdown / knockout detection. Therefore, the unstable GFP (uGFP) was created as the destabilized version reporter. The uGFP is best used for the time course induction and dose-response kinetics, and for the fast response to knockdown or knockout.
Lentivirus express the unstable GFP:
Gentarget developed the expression lentivirus for the destabilized GFP (uGFP) (click to see its sequence) fluorescent protein (475nm/510-525nm, Ex/Em), driven by an optional inducible CMV promoter (TetCMV) with different antibiotic selection marker. See core lentivector scheme below.
Lentivirus are provided as in two formats:
- Crude Lentivirus: 200ul /vial in DMEM containing 10 x Polybrene;
- Concentrated lentivirus: 200ul PBS solution which is better for hard-to-transduced cell types or for serum-free culture.
