About EGR1 Promoter Pathway:
EGR-1 (Early growth response protein 1) is a mammalian zinc finger transcription factor. The full human EGR1 promoter sequence contains regulatory elements including two Sp1 sites, an AP1 site, two cAMP response elements and an Egr-1 binding site, five serum response elements (SREs). This promoter is activated by ionizing radiation, demonstrating radiation-induced upregulation. It is also up-regulated in response to growth factors or stress that activated by the MAP kinase kinase MEK1. Therefore, dependent on specific cell type, the activation of EGR1 promoter involves multiple signal transduction pathway.
The feature that the EGR1 promoter is induced by ionizing radiation, various mitogenic stimuli, malignant transformation signals, makes it a useful in vivo tool for cancer research.
Monitor EGR1 Promoter Pathway:
GenTarget developed a set of EGR1 reporting lentivirus to monitor EGR1- promoter signaling pathway. Those reporting lentivirus has a luminescent report (Luciferase, Renilla Luc) or a fluorescent report (GFP, RFP) or a secreted SEAP report, under the full sequence of human EGR1 promoter. Those EGR1 reporter are used to measure all pathway signals that activate EGR1 transcription factor. Those reporter monitor both increases and decreases in the activity of EGR1 pathway, and determine the effect of various treatments, such as gene knockdown, over-expression, and chemical compounds on EGR1 activity.
The luciferase signal can be measured via Luciferase assay. The fluorescent reporter can be detected via its fluorescent signal. The SEAP secreted into cell culture supernatant, which allows to determine reporter activity without disturbing the cells, does not require the preparation of cell lysates and can be used for kinetic studies.
How to Use EGR1 Promoter Report Lentivirus:
Those reporting lentivirus contains a constitutively expressed fluorescent selection marker or an antibiotic selection marker under the RSV promoter, which makes it easier to select the stably infected signal reporting cells (to generate pathway specific sensor cell lines), or provides internal reference for virus transduction efficiency when a fluorescent marker is under the RSV promoter. The premade, ready-to-use reporter lentivirus provides a much easier tool to monitor the activity of EGR1-Promoter signaling pathways in virtually any mammalian cell type. It also allows to generate your own reporting cell line in your desired cell type for study or screen of pathway specific gene-knockdown, over-expression, or chemical / drug/protein treatment in the cell based assay.
