Description
Pre-made lentivirus express SEAP reporter under the SFFV promoter. It can be used to generate SEAP reporter cell line, and to check SFFV promoter strength in your given cell types, and to other application.
The spleen focus-forming virus (SFFV) promoter is derived from the SFFV retrovirus and is known for its strong and constitutive activity in a wide range of cell types. It is often used in gene therapy constructs to drive high levels of gene expression in hematopoietic stem cells.
The SEAP (Secreted Embryonic Alkaline Phosphatase) is commonly used as a powerful reporter gene whose signal can be detected via chemiluminescent reporter assay based on dioxetane CSPD (chloro-5-substituted adamantyl-1,2-dioxetane phosphate), and provides a convenient and highly sensitive method for the quantitation of transcriptional activity. SEAP secreted into cell culture supernatant and therefore offers many advantages over intracellular reporters, like luciferase. It allows to determine reporter activity without disturbing the cells, does not require the preparation of cell lysates and can be used for kinetic studies.
This lentivirus also contain the Puromycin selection under the constitutive RSV promoter for antibiotic selection of the transduced cells. See Product Manual for details (.pdf).
Amount: 200ul/per vial, at titer of 1 x 108 IFU/ml in PBS (Premixed with 10x polybrene, 60ug/ml)
